“In this project, Country Day students collaborate on primary scientific research. We started by 3D printing a dynamic physical model of a protein backbone using the open source code provided by the authors of an interesting paper who have been happy to help us learn their techniques. We are now extending their work as we aim to print a dynamic model of an entire protein: insulin. Pictured in the background of this page, insulin is a small hormone involved in the regulation of sugar in the body and will require the modeling of many other residues than the alanine described in the original work. To date, we have successfully printed accurate models of the disulfide bond between two cysteine residues (depicted in yellow below) and the proline residue (which contains a unique ring-structure, middle-right in the image below). In the coming years, students will continue to build on this body of work as they gain invaluable experience in the exciting field of scientific research.” - Tom Collins, Project Creator & Brittany Lott, Project Advisor

In addition to the much-publicized environmental impact of CO2 emission by air traffic, aviation particulate emission also deserves attention. The abundant ultrafine particles in the aviation exhaust with diameters less than 100 nm may penetrate deep into the human respiratory system and cause adverse health effects. Here, we quantified the detailed aviation particle number emission from Zurich Airport and evaluated its influences on the annual mean particle number concentrations in the surrounding communities. The actual flight trajectory data were utilized for the first time to develop an emission inventory with high spatial resolution. The estimated total particle number emission was in the magnitude of 1024 particles per year. The annual mean particle mass concentrations in the nearby communities were increased by about 0.1 μg m–3 due to the aviation emission, equivalent to about 1% of the background concentration. However, the particle number concentration could be increased by a factor of 2–10 of the background level (104 cm–3) for nearby communities. Further studies are required to investigate the health effects of the increased particle number concentration and to evaluate whether the regulation based on the mass concentration is still sufficient for the air quality near airports.

g2nb/Single-sample-GSEA-projection-ssGSEA-
Single-sample GSEA projection (ssGSEA) Background Traditional gene set enrichment analysis assesses the differential coordinate up- or down-regulation of a biological process or pathway between groups of samples belonging to two phenotypes. The ability to assess that enrichment in individual samples, especially independently of pre-assigned phenotype labels, provides the opportunity to analyze transcription data at a higher level, by using gene sets/pathways instead of genes, resulting in a much more biologically interpretable set of features. Single-sample Gene Set Enrichment Analysis (ssGSEA) Projection accomplishes this. ssGSEA projects a single sample’s gene expression profile from the space of single genes onto the space of gene sets. It does this via the ssGSEA enrichment score, which represents the degree to which the genes in a particular gene set are coordinately up- or down- regulated within a sample. Any supervised or unsupervised machine learning technique or other statistical analysis can then be applied to the resulting projected dataset. The benefit is that the ssGSEA projection transforms the data to a higher-level (pathways instead of genes) space representing a more biologically interpretable set of features on which analytic methods can be applied. Another benefit of ssGSEA projection is dimensionality reduction. Typically the number of gene sets employed in the enrichment analysis is substantially smaller than the number of genes targeted by a gene expression assay, and they are more robust and less noisy, resulting in significant benefits for downstream analysis.

This project was created by the Fall 2017 Mapping Controversies class in “Personal data protection regulation: global social, legal and technical challenges” at Sciences Po, in Paris, France. The focus of our investigation was data protection and privacy regulations. The class, composed of students, represents diverse backgrounds spanning from France, Argentina, United States, Israel, Hungary, Russia and Canada. In the course, the group first investigated scientific literature concerning data protection and privacy regulations, casting a wide net in our investigation and gathering a list of key policies, terms, laws, and actors involved in the controversy at hand. Afterwards, the group investigated press articles, reviewed web materials, looked through social media, and reviewed legal documents concerning the controversy. This included, for example, a review of key actors on Twitter and led to a map of these actors’ networks on Twitter. We were interested in who was discussing these issues, how they interacted, how the technology was advancing to address or create these controversies and how policy makers are reacting. The following website shows off our investigation into the multi-faceted controversies regarding data protection and privacy regulation. We hope you enjoy.
